| Тойм: | <p>Juego de datos de electrofisiología para construir las Figuras 6D y 6E del manuscrito "A filopodia-based dendritic mechanosensory compartment in CSF-contacting neurons" https://doi.org/10.64898/2026.04.09.713694.</p> <p>The details of the analysis are explained in the “METHOD DETAILS” section of the above mentioned article. Briefly, we prepared transverse or 45°-angled lumbar spinal cord slices (300 µm thick) from GATA-3 eGFP mice and performed patch-clamp recordings from CSF-cNs. We used the voltage-clamp configuration to record mechanically evoked currents, and the cell-attached mode to assess the impact of these stimuli on neuronal excitability. </p> <p>For mechanical stimulation we positioned a pipette with Luigs & Neumann micromanipulators to stimulate the ApPr or soma with a 2 µm (or 4 µm) 'step' producing an indentation (push) on the desired compartment. We acquired data at 20 kHz (low-pass filtered at 2.9 kHz) using a Heka Elektronik EPC 10 USB Patch Clamp Amplifier (RRID:SCR_018399) and Patchmaster software (RRID:SCR_000034). </p> <p>We visualized eGFP and Alexa 594 using a dual LED light source (Optoled; Cairn Research) and an Andor Ixon EM CCD camera (Oxford Instruments) with appropriate filter sets (Chroma, USA). We applied the PKD2L1 channel inhibitor dibucaine (Sigma-Aldrich Cat# D0638) by puffing a 200 µM solution in extracellular Ringer for 30 s. We analyzed electrophysiological data in IGOR Pro (WaveMetrics, USA; RRID:SCR_000325) using NeuroMatic (RRID:SCR_004186), TaroTools and custom routines.</p> |
